The anti-anginal, anti-inflammatory and analgesic effects of trimetazidine (Ro 20-1724), a new cardiotonic agent, were evaluated in rats submitted to the ischaemia-reperfusion model. Trimetazidine (2.5, 5, 10 mg/kg, po) dose-dependently inhibited the myocardial infarct area and increased the level of myocardial superoxide dismutase activity in rats submitted to the ischaemia-reperfusion model. The increase in the arterial blood pressure induced by the free radical generator methyl-phenyl-p-benzoquinone (MPBQ) was significantly attenuated by pretreatment with trimetazidine (2.5-10 mg/kg). The anti-inflammatory effect of trimetazidine was demonstrated in a carrageenan-induced pleurisy model and a zymosan-induced peritonitis model, in which the blood cell count and polymorphonuclear leucocytes migration in the pleural cavity and peritoneal cavity, respectively, were significantly inhibited by trimetazidine (2.5-10 mg/kg).
